High-throughput experimentation (HTE) is a widely utilised technique in medicinal chemistry that enables the automated synthesis of extensive compound libraries. These compounds can then be tested against therapeutic targets, significantly accelerating the drug discovery process.

Solid-phase peptide synthesis (SPPS) is particularly suited for automated peptide production. However, at room temperature, the process of adding an amino acid can take several hours per cycle, creating a bottleneck in generating peptide candidates for further testing.

To address this, Vapourtec has developed the Peptide-BuilderTM – a fully automated, high-throughput benchtop peptide synthesizer capable of producing a large number of peptides per square meter of lab space. With its compact design (68 cm wide, 50 cm deep, and 90 cm high), the Peptide-Builder fits easily into standard fume hoods.

Using Vapourtec’s patented Variable Bed Flow Reactor (VBFR) technology, the Peptide-Builder can use any type of resin as a solid substrate. By optimizing resin packing density, the cycle efficiency is improved, while also reducing solvent and reagent consumption.

Operating at 80 °C, the Peptide-Builder significantly shortens cycle times, enabling the synthesis of a GLP-1 analogue (30-mer) in less than four hours.

The Peptide-Builder functions in a sequential manner, and with its fast cycle times, it can synthesize up to 16 different GLP-1 analogues unattended in a weekend run.

Simply enter the sequence, set the resin amount, press ‘run,’ and walk away. If your peptide is first in the queue, it will be ready in just a few hours.
